Genotoxicity of iron sulfate in mice and possible protective effect of Olive oil

Abstract
Sixty four Swiss albino male mice (Mus musculus), were used for assessment of iron sulphate acute (after 24 and 48 hrs) and chronic (six weeks) genotoxicity using chromosomal analysis, mitotic index and RAPD-PCR assay. Five experimental groups were used, first group acts as a control without any treatment. The second group received 75 µl EVOO in diet for one day before samples collection. The third group received 61.2 mg/kg b.wt iron sulfate in diet which is 1/5 of oral LD50). The forth group received the same previous doses of EVOO and iron sulfate at the same time in diet for 24 hrs. The fifth group received EVOO with previous dose for one week then gave iron sulfate with previous dose for 24 hrs.
The acute and chronic iron genotoxic is definite, through induction of structural and numerical chromosomal aberrations, lowering the mitotic index and DNA fragmentation by RAPD-PCR. The protection of EVOO against iron sulfate treatment is low.
