Abstract
The current investigation sought to assess the efficacy of a nanopolymer derived from the small intestine submucosa (NSIS) for the regeneration and recovery of the neurectomized sciatic nerve. Thirty-two adult male rats were randomly assigned to two equal groups (n=16) following induction of left sciatic nerve neurotmesis with a 0.5 cm defect: in the first group, the transected nerves were coaptated using a silicone tube loaded with carbon nanotubes (CNTs group); in the second group, the transected nerves were coaptated with a silicone tube containing a nanopolymer of SIS (NSIS group).
Clinically, the animals of the NSIS group demonstrated a significant improvement (P ≤ 0.05) in motor and sensory functions with less tibialis anterior and gastrocnemius muscle atrophy when compared to the CNTs group during periods of assessment. The histopathological findings suggested that the disparities in nerve regeneration between the CNTs and NSIS groups may be ascribed to differences in Schwann cell quantity, diminished collagen fibers at the conduit site, enhanced nerve fiber orientation, and a reduction in nerve degeneration within the NSIS group. Additionally, the qRT-PCR results indicated that the NSIS considerably stimulated neurotrophics, including basic fibroblast growth factor (bFGF), ciliary neurotrophic factor (CNTF), and brain-derived neurotrophic factor (BDNF). In conclusion, NSIS provided a favorable microenvironment supporting regeneration and recovery of the neurotomized sciatic nerve.

Key words: Keywords: Carbon Nanotubes, Nanopolymer of SIS, Growth Factor, Sciatic Nerve.
