Abstract
Background: The bovine fatty acid synthase (FASN) gene encodes a multifunctional enzyme that carries out de novo fatty acid synthesis in the mammary gland. Its main product is palmitic acid (C16:0), and this activity helps shape the milk fat profile. However, information regarding the association of FASN polymorphisms with milk production traits in Egyptian Holstein–Friesian cattle remains limited.
Method: Samples of blood and milk were obtained from 70 Holstein-Friesian dairy cattle. DNA was extracted from blood samples for the PCR amplification of a 353 bp fragment of the FASN gene. The HhaI restriction enzyme was employed for RFLP investigations focusing on SNP g.17924A>G located in exon 34 of FASN gene, followed by direct DNA sequencing. The composition of milk was analyzed by Lactoscan milk analyzer.
Results: RFLP investigation revealed polymorphism with three genotypes: AA, AG, and GG. The GG genotype exhibited a significant increase in milk yield, fat, protein, and lactose percentages. Direct DNA sequencing confirmed the g.17924A>G non-synonymous SNP that causes a threonine-to-alanine amino acid substitution, in addition to a C/T synonymous SNP.
Conclusion: The current finding indicates that genetic polymorphism in the FASN gene was associated with milk yield and composition traits in Holstein-Friesian dairy cows. The favorable performance of the GG genotype suggests that the g.17924A>G polymorphism could serve as a promising candidate marker for incorporation into marker-assisted selection (MAS) strategies to improve milk yield and composition traits and overall dairy breeding efficiency.

Key words: FASN polymorphism, Holstein-Friesian cows, PCR-RFLP, sequencing, milk composition.
